Hepcidin-25是一種由25個(gè)氨基酸組成的肽類激素,在其肽鏈中存在著一條金屬結(jié)合基序,并在脊椎動(dòng)物鐵代謝調(diào)控中發(fā)揮核心作用
1。作為鐵穩(wěn)態(tài)的"主調(diào)控因子",它通過抑制腸道鐵吸收和巨噬細(xì)胞鐵釋放來調(diào)控循環(huán)鐵的水平
2。Hepcidin-25常常作為疾病模型中的分析指標(biāo)或者生物標(biāo)志分子。例如在健康狀態(tài)下,大鼠血漿中Hepcidin-25的平均濃度為63.0 ng/mL,而在脂多糖刺激后顯著升高至218.2 ng/mL,表明Hepcidin-25與炎癥反應(yīng)高度相關(guān)
3。在心血管領(lǐng)域,研究發(fā)現(xiàn)Hepcidin-25與動(dòng)脈僵硬度存在關(guān)聯(lián)
4。Hepcidin-25還可調(diào)節(jié)鐵代謝,Hepcidin-25與其受體鐵輸出蛋白結(jié)合后,可抑制腸道鐵吸收以及肝細(xì)胞和巨噬細(xì)胞的鐵外排。檢測(cè)Hepcidin-25可以預(yù)測(cè)機(jī)體的鐵代謝狀態(tài),并有助于研究貧血和某些以貧血為特征的動(dòng)物疾病模型,例如慢性腎臟。–KD)、糖尿病相關(guān)的慢性腎病、炎癥性腸病血色病等動(dòng)物模型
5。Hepcidin-25的升高還可用于監(jiān)測(cè)腫瘤的發(fā)生發(fā)展,例如Hepcidin-25與肝細(xì)胞癌的腎臟轉(zhuǎn)移有關(guān)
6,并且Hepcidin-25還可作為非侵入性乳腺癌預(yù)測(cè)工具。目前針對(duì)Hepcidin-25的檢測(cè)手段包括LC-MS/MS法、ELISA檢測(cè)試劑盒等技術(shù)。Hepcidin-25還具有其它的生物學(xué)應(yīng)用例如抗菌,有研究表明Hepcidin-25的水平在細(xì)菌感染后會(huì)明顯上調(diào),表明其與機(jī)體的主動(dòng)防御有關(guān)。而在體外實(shí)驗(yàn)中,Hepcidin-25及其衍生物Hepcidin-20對(duì)多種病原微生物表現(xiàn)出抑制活性
