ISRIB(Integrated Stress Response Inhibitor,AbMole,M9242)是一種小分子化合物,能夠特異性抑制整合應(yīng)激反應(yīng)(ISR)的激活。ISR是細(xì)胞在應(yīng)激條件下(如缺氧、病毒感染等)通過(guò)磷酸化真核翻譯起始因子2α(eIF2α)來(lái)調(diào)控蛋白質(zhì)合成的保守通路[1]。磷酸化的eIF2α會(huì)抑制鳥苷酸交換因子eIF2B的活性,從而減緩蛋白質(zhì)合成[2]。ISRIB通過(guò)結(jié)合eIF2B的特定位點(diǎn),逆轉(zhuǎn)eIF2α磷酸化對(duì)eIF2B的抑制,恢復(fù)蛋白質(zhì)翻譯[3]。
ISRIB(Integrated Stress Response Inhibitor,AbMole,M9242)通過(guò)調(diào)控整合應(yīng)激反應(yīng)表現(xiàn)出多種生物學(xué)效應(yīng):SRIB能減輕β淀粉樣蛋白(Amyloid β Protein)誘導(dǎo)的神經(jīng)元死亡,但不影響β淀粉樣蛋白的生成[4]。在膠質(zhì)母細(xì)胞瘤(GB)細(xì)胞中,ISRIB可增強(qiáng)細(xì)胞對(duì)Temozolomide(TMZ) 和低氧誘導(dǎo)的凋亡的敏感性[5]。ISRIB在小鼠模型中,能改善慢性應(yīng)激(CRS)誘導(dǎo)的抑郁樣行為,表現(xiàn)為蔗糖偏好增加和強(qiáng)迫游泳試驗(yàn)(FST)中不動(dòng)時(shí)間減少[6]。
此外,ISRIB可減輕腦損傷后的認(rèn)知功能衰退,并增強(qiáng)健康小鼠的認(rèn)知能力[7]。ISRIB在結(jié)核分枝桿菌感染的小鼠模型中,通過(guò)抑制PKR介導(dǎo)的eIF2α磷酸化,阻斷了肺肉芽腫壞死的發(fā)生[8]。這些發(fā)現(xiàn)表明,ISRIB可作為調(diào)控蛋白質(zhì)穩(wěn)態(tài)的工具分子,為理解ISR在腫瘤、免疫和神經(jīng)功能中的作用提供了幫助。
